About Bortezomib

A proteasome inhibitor that revolutionized multiple myeloma treatment. Bortezomib or Velcade treats multiple myeloma by inhibiting the proteasome, causing cancer cells to accumulate damaged proteins and die. Before bortezomib, multiple myeloma had a short median survival. With bortezomib and newer therapies, survival has improved substantially. The drug can cause peripheral neuropathy. Subcutaneous administration reduces this side effect compared to intravenous. Bortezomib demonstrated that targeting protein degradation could treat cancer.
